У последњем периоду Др Ана Рилак Симовић је ангажована на истраживању у вези са дизајном и синтезом нових комплекса рутенијума и злата као потенцијалних антитуморских агенаса. Синтетисани су и окарактерисани нови комплекси злата(III) са хлорофенил терпиридин лигандом, као и комплекси рутенијум(II/III) са лигандима Шифове базе и полипиридил лигандима. Део истраживања односи се на испитивање кинетике и механизма реакције супституције са биомолекулима као што су гуанозин-5′-монофосфат, L-хистидин и L-метионин помоћу UV-Vis спектроскопије. Други део истраживања односи се на испитивање интеракција комплекса са ДНК и хуманим серум албумином. Афинитет везивања комплекса за ДНК проучаван је UV-Vis и флуоресцентном спектроскопијом, као и вискозиметријом. Такође су испитиване компетитивне интеракције везивања са интеркалационим агенсом етидијум бромидом, као и са Hoechst 33258, који се везује за мањи жлеб. Поред тога, испитиване су компетитивне интеракције комплекса са хуманим серум албумином у присуству маркера ибупрофена и езонаY. Трећи део истраживања односи се на in vitro и in vivo испитивања антитуморске активности новосинтетисаних комплекса прелазних метала.

 

In the last period, Dr. Ana Rilak Simović was engaged in the research related to the design and synthesis of new ruthenium and gold complexes as potential antitumor agents. New gold(III) complexes with a chlorophenyl terpyridine ligand, as well as ruthenium(II/III) complexes with Schiff base ligands and polypyridyl ligands, were synthesized and characterized. Part of the research refers to the investigation of the kinetics and mechanism of substitution reactions with biomolecules such as guanosine-5′-monophosphate, L-histidine and L-methionine, by UV-Vis spectroscopy. The second part of the research is related to the examination of the interactions of the complex with DNA and human serum albumin. The binding affinity of the complex to DNA was studied by UV-Vis, fluorescence spectroscopy and viscometry. Competitive binding interactions with the intercalating agent ethidium bromide, as well as with Hoechst 33258, which binds to the minor groove, were also investigated. Additionally, the competitive interactions of the complexes with human serum albumin were investigated in the presence of markers ibuprofen and ezone Y. The third part of the research refers to the examination of the in vitro and in vivo antitumor activity of the newly synthesized transition metal complexes.
